    RNA-Seq sample prep...

    Hello-

    Long time NGS researcher...first time RNA-Seq. I'm trying to figure out how to do sample prep. There are various kits available, and they all claim to be the latest and greatest. Obviously they're not all the greatest. Before spending money I'm trying to gauge what seems to be working well in the community, and which appear to be not so good. So, what are you using and how well is it working? Additionally, any insights comparing two (or more) methods would be greatly appreciated. Thanks!

    I've been making RNA-Seq libraries for three years now, basically following the same protocol as one of the first papers (Marioni et al 2008) with a few modifications (e.g RNA fragmentation on Covaris). It works well for me and the costs/library are a fraction of the commercial kits as I buy all the enzymes/reagents from the individual vendors in bulk.
