    Truseq RNA kit for making DNA library

    I want to make 500bp paired-end library from genomic DNA. I already have the Truseq RNA kit. I only have two samples and don't want to order the expensive Truseq DNA kit. I check the manual and wandering maybe we can use the RNA kit from end repair step after sonicating. Does anyone try this?

    Although your post is from long ago, I was wondering how you finally ended up. I am in a similar situation now.
    I read that the adaptors in the RNA and DNA TruSeq kit are the same, though the concentrations are not.

      It works totally fine. I've done it. The two kits a indistinguishable once you have clean dsDNA. The primers and reaction mixes are at the same concentrations.

        We did use RNA kit for all of our RNA/DNA cases. They were doing well and we could happily maximize the use of the RNA kit.

          Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
