I'm using Masterpure's Gram Positive DNA purification kit, trying to isolate
DNA from Propionibacterium acnes. Each attempt I get really graphs from the nanodrop.
I defnitely have DNA in my samples, because i'll have a high absorbance at 260, but sometimes the absorbance is even HIGHER at 230 nm.
I've tried doing an extra purification step, where I repeat the steps where you add isopropanol, spin at 4C, remove the isopropanol, wash with ethanol, and resuspend in 35ul TE.
Has anyone experienced DNA isolation where they had a high 230 absorbance, and what did you do to fix it?
