    Agilent SureSelect XT2 pre-capture library construction

    Hi all,
    I'm wondering if anyone has worked with the Agilent SureSelect XT2 protocol which features pre-capture library pooling. I am attempting this protocol for the first time and my individual pre-capture library yields were extremely low. I've previously worked with the Human All Exome protocol (no pre-capture library pooling) and have had no issues with my pre-capture library construction. I tried to amplify the pre-capture libraries here with the recommended 5 cycles of PCR and did not get the yields I expected. I was hoping someone else has had experience with this protocol and how many cycles of PCR were needed for their pre-capture libraries. Thanks in advance!!

    We are looking at this pool as well, but have not yet made the decision to dive in (har-har). Has anyone had significant experience with pre-capture pooling?
