    Choosing between Nextera and Nextera XT

    Hi all!

    we are going to sequence an important amount of yeast strains (Saccharomyces cerevisiae), and we are doubting in between using nextera or nextera xt to prepare the libraries.
    Does anyone have any experience with these two kits (or one) and their efficiency when used yeast's genome?
    How much does the size of the genome affect the performance of the kit? Any recommendations?

    Any help is going to be hugely appreciated!

    We only sequence bacteria genomes. But maybe our experience on Nextera and Nextera XT may help you choose the kits. We started by using Nextera sample prep and then switch to XT. Main reason is because of the significant price difference. Second, we dont find the sequencing quality much different to Nextera.

    The starting material for Nextera is 50ng and for Xt is 1ng. So we were quite nervous in the beginning. But after 10 MiSeq runs, we are quite satisfied with XT sample prep, especially on a 250bp PE run.
