For the reverse crosslinking step in ChIP, we use 4uL of 5M NaCl for every 100uL of DNA and incubation occurs at 66oC for overnight.
Just wondering if anyone has tested whether the time used for the crosslink reversal step affects the yield of the DNA recovery? i.e. some protocol says 4 hours and some says 16hrs, which is better?
