    library prepare for long targets

    Hi All,

    I have trouble for library preparation. I sheared my gDNA with sonic system and do end repair,dA tailing, and ligate adapter in one side and another barcode adapter in another side, but when I do PCR with primer designed on adapters (here I call adapter primer and barcode primer), but I only get the PCR products with adapter primer in both sides! The expected products should have the adapter primer and barcode primer in each side.

    I don't know why, can someone help me top figure out the problems?？
