    Mitochondrial Genome Sequencing

    Hi All,

    I am trying to determine which library prep kit to use for whole mitochondrial genome sequencing (assuming mitochondrial genomes size of ~17,000bp).

    I'm currently thinking that the Illumina TruSeq DNA PCR-Free Sample Preparation Kit is our best option, but am curious about whether other kits might be better or if any of you have mitochondrial genome sequencing experience using other kits.

    We are using Nextera XT, works fine with plant mitogenomes.

      Are you purifying the mtDNA before starting library preps?

        nope, since we didn't enrich mtDNA.
