Hello,
I'm preparing DNA for library prep for Illumina paired end sequencing using a Diagenode Bioruptor Pico for fragmentation. I would like to shear the DNA to approx. 550 bp. The genome size of my sample (land snail) is 300 Mb.
I ran my test sample at 12" on 90" off for 7 cycles (after 3 cycles the sample was vortexed) and became an average size of 544 bp ( which was pretty close), but the peak is definitely too broad (see attachment). Does anybody know how to reduce peak width? Or does anybody have some experience with shearing dna of land snails?
Test Sample: 100 ul in AE at 20 ng/ul in 0.65ml tubes.
I would be very thankful for any input!
