    Weird bioanalyzer of stranded RNA library

    What could be the reason for this bioanalyzer results for a total RNA stranded lib prep? I have been using the low mammalian input kit from clontech.

    Could you post a higher-resolution version of this image and an electropherogram of the template RNA itself? This looks like it might be the result of degraded RNA.

      I see the same in my positive control which is suppose to be RNA of good quality.

        They could both be degraded if you had RNase contamination at some step. How do the RNAs look on the Bioanalyzer?

          What organism did the starting RNA material come from? A "spiky" RNA-seq library trace such as this often indicates the presence of high abundance transcripts. It is possible that your ribosomal depletion step was not very efficient.
