After obtaining the amplified adapter-ligated cDNA fragments, the Illumina protocol suggests the cloning of 4% of the volume of the library into a sequencing vector so in order to sequence the individual colonies. I have size selected 150-350 bps cDNA fragments. What will conventional sequencing tell me? The protocol says u can verify that insert sequences are from the genomic source DNA. But I don't get how. Can anyone please explain it to me?
