    Metagenome analysis from low biomass samples

    I have access to a sample that we hope to run a microbial ecological study on. The problem is that each sample only has about one picogram of DNA.

    What are the best molecular biology and/or bioinformatics techniques to look into to maximize the chance I can develop RNA-sequencing analysis techniques from such low biomass samples?

    I do not think that there is any hyper sensitive method yet to preprare RNA-Seq library from 1 pg RNA.

      Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.


      The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.
