library clean-up using AMPure XP beads

Hi all,

I'm having some problem when performing library clean-up, I found when I reduced the bead ration from the recommended 0.8X twice to 0.7X twice, I can almost remove all the adapter dimers in my library, but when I analyzed the sequencing data from such library, I found the mapping rate reduced from original 40% to less than 10%, I wondered is this because I reduced the bead ratio, which cause the loss of a good proportion of my fragment?
