We just ran a PTP with a new lot of sequencing reagents that totally failed (0 passed filter wells, no control or library sequences. It looks like Roche has already moved on to a new lot so I am wondering if they found problems with this one. The lot in question is 93884720. Has anyone else experienced poor results with this lot?
The only other thing I can think of is that the PTP itself had a different look to it (more sheen than normal, I remember thinking when loading it that it looked different than the other 30 that I have seen.) Also, the DNA bead layer seemed to slough off a lot of beads when the supernatant was pulled off. Mistakenly, I didn't think to count these beads to see if any actually got into wells.
Given that no control beads sequenced, I don't think this is an issue of forgotten primers or anything like that.
Any input is extremely welcome. We have never seen this before and are wary of running the same beads again today.
