  • Roche new software version (2.7) from this Spring

    Hey all,

I was wondering how the 2.7 software was received by the sequencing community? We are thinking about installing it pretty soon, but have heard that it was buggy in the early stages and some people were unhappy. Any and all experience with the new software that came out this spring would be much appreciated. Running the GS Junior BTW

I understood it was a GS Junior release only, and wasn't planned as a general release.

...and it would seem I'm the only GSJ user on the forums

I don't think they sold many of them. I reckon they would have done a LOT better if the yield was just a bit higher! As it stands, it is just a little bit too low to be useful for bacterial genome assemblies (except for small 1-2Mbp ones).

That's true, we only use ours for 16s sequencing and pooled libraries of many samples, but it works well enough for that. I guess I will keep my fingers crossed that the one other user installed the software and reads this!

I have the 2.7 software and I'm using it for data analysis. I have a FLX, so the instrument is of course still at 2.6 and I'm waiting for 2.8 for it. I got the 2.7 for data analysis mainly for the changes in the AVA software. I find those changes help me out a lot by getting rid of a lot of the spurious reads instead of making them consensus reads. Otherwise I can't say much about it.

                Strategies for Sequencing Challenging Samples
                by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
