  • Sequencing of E. coli strains

    I currently have a inquiry for the sequencing of 2 E. coli strains in hand. They want to use a 20kb paired end library for the sequencing task. I always thought that 3-8kb would be sufficient for the sequencing of prokaryotes ...and that 20kb libraries are used for eukaryotic sequencing tasks?

    What is your opinion? What is the best way to sequence a prokaryotic genome?

    20kb seems (is) overkill. I would stick to 8kb unless you have indications that there is a 15kb (or so) repeat in the genome...
