  • SureSelect and Rapid MID Libraries

    Hello Everyone,

    I have recently used a custom Agilent SureSelect bait library to enrich several Illumina multiplexed libraries (combined and loaded on a single lane). I have left over bait library and would like to use it to do the same thing with Rapid 454 MID libraries. I recently found out that additional Rapid MID adaptors can now be purchased from IDT. These MID adaptors are delivered as two oligos that must be annealed into a double-stranded adaptor prior to library preparation.

    I wanted to know.....

    1)Can the separate oligos (not annealed) be used for Linker Mediated PCR (LM-PCR) to amplify sample libraries pre and post hybridization with the SureSelect baits?

    2)Can the separate oligos (not annealed) be used as blocking oligos in the hybridization?

    I think the oligos contain a FITC and/or two or three Inosine-molecules, so I wouldn't try both.

      Thanks Zaag for the information. I've decided to use the Nextera DNA Titanium-compatible library kit with their additional bar code kit. The sequences of their adapters/primers are available so it should be easy to design oligos for enrichment PCR and blocking oligos for the hybridization.
