  • Sequencing reagents lot 93801720

I was wondering if anyone had experienced problems with the above sequencing reagents lot?

We performed a run a couple of days ago which has not generated a single read. The Raw wells for this 4 region run was around 1.3 million but both the sample key pass and control key pass were both 0. It maybe a problem with the instrument or technician but as this was the first run we’ve performed using this lot I thought I’d ask if anyone else was having problems with it.

If there are 0 wells for control beads, then there is likely one of 2 problems:
    1. the control beads weren't added (likely)
    2. there is a problem with the instrument (only if you didn't get anything from the sample beads too

I would start by rerunning and making sure the control beads were added in the correct volume. If you still don't get any results for the control beads, then 454 needs to come look at your instrument.

Was the sequence primer annealed? (Edit: you would of course still have control beads with a sequence primer so stupid question on a mondaymorning)

We used this lot successfully on 4 runs in October.

Essential Discoveries and Tools in Epitranscriptomics
          by seqadmin

The field of epigenetics has traditionally concentrated more on DNA and how changes like methylation and phosphorylation of histones impact gene expression and regulation. However, our increased understanding of RNA modifications and their importance in cellular processes has led to a rise in epitranscriptomics research. “Epitranscriptomics brings together the concepts of epigenetics and gene expression,” explained Adrien Leger, PhD, Principal Research Scientist...

          Current Approaches to Protein Sequencing
          by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...
