I assume you are using CASAVA version 1.8.Originally posted by cjose View PostI just started using CASAVA and currently demultiplexing now. Can anyone explain what exactly is demultiplexing? Having a trouble understanding the purpose of it. Thanks in advance.
If you do not have "multiplexed" samples then this version of casava treats samples being multiplex of "1". When you are "de-multiplexing", the process is converting the "bcl" files (primary sequencing output) to actual FASTQ sequence files. If you do have real multiplexed samples then it will also split the reads into bins based on the "tag" info you provide in the SampleSheet file.

Tumors contain complex mixtures of cell populations that evolve in response to treatment and host-specific factors. Tissue biopsies provide only static snapshots of a tumor’s state. But what if we could follow that evolution over time using a minimally invasive technique?
Liquid biopsies can detect tumor-derived material circulating in the bloodstream, providing a way to study treatment response, detect emerging resistance, and discover new biomarkers. Circulating...-
