    Library durability

    I am using the Illumina kit TruSeq NANO DNA library prep to obtain my libraries before sequencing capture. However, for external reasons, I will have several (up to 6) months between the two process. I saw that in the protocol appears that the maximum expiration date after ligate adapters is about 7 days and I was wondering if there's any other process to keep them longer, and/or if anyone has tested them before.

    If you have done PCR after ligation, cleaned library can be stored for years. Note that some library fragments may not be amplifiable (after capture) but still would hybridise to capture probes and may reduce capture efficiency. Though I think that 6 months storage at -20C should not have large effect.
