  • is is possible?

We are planing to move to Solexa from 454, the problem is if it is possible to get a good assemble from only Solexa data for the human Mitochondrial DNA(17Kb, circular, no repetitive element)?

I have seen good assembly for plant mitochondrial DNA using 35bp reads. The fact that there's no repetitive element makes it easier.

Do you have the reference paper?

Oops...my mistake. What I saw is chloroplast DNA (~160kb) not mitochondrial DNA. I think it's not published yet.

I asked some experts, it seems highly possible, although nobody publish it yet.

We have sequenced human mito DNA on the Illumina and get very good results.

              Hi Bruce,

What's your software to do the assembling?

                Strategies for Sequencing Challenging Samples
                by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
