  • MiSeq run failed (no usable signal found in the images)

    I had a run on miseq using v3 kit and within an hour the run stopped with the error "no usable signal found in the images it is possible that clustering has failed". I have read that hardware could be an issue but I was just wondering can this happen with wrong library quantification? I did quantified library using Picogreen assay kit and diluted the libraries, however now I feel suspicious about the kit's accuracy.

    So I have 2 questions:
    1) is it possible that clustering failed that early if the library was not diluted correctly.

    2) since clustering didn't even start, what would be the status of that flowcell? I do have an old expired v3 kit I wanted to try but do not have a flowcell for it. So my question is, can this flowcell be reused with another kit since no clustering was started?

    just happened to me
    JUST HAPPENED, just yesterday
    I am waiting for an explanation from the IlluminaTech

    I quantified my sample with Qubit 1X HS DNA, I also checked with BioAnalyzer for size distribution. Everything was OK. I also re-checked the pooled concentration, it was expected, based on the average library size.

    Mine was stopped just before two hours.
    the error message was "ThroughFocusScan CoarseFocus failed to find focus"

      can you attach some of the first few images from the cycles? I expect its overclustered and you can tell from that

        It is likely to be overclustering. We encountered similar problems before, need to re-quantified (qPCR) the libraries.

          Illumina did not really give me an answer,
          they (kind of) insisted that it was the library fault.
          I re-run with a new cartridge (v3),
          with the exact same library.
          It was fine.
