  • SMRT Tools HiFiViral_SARS-CoV-2_M13barcodes?

Hello, I'm a bioinformatics analyst trying to run some coronavirus samples through the HiFiViral SARS-CoV-2 Analysis application, a part of SMRT Tools. I need to demultiplex the ccs.bam files I have (see "Preparing Input Data, pg. 44: https://www.pacb.com/wp-content/uplo...uide_v10.2.pdf).

The guide states to "Provide the proper barcode sequences: HiFiViral_SARS-CoV-2_M13barcodes." Okay, neat, sounds super easy, barely an inconvenience.

I can't find those anywhere. Not on GitHub, not in the project metadata, not on forums. I emailed PacBio and didn't get a response. Does anyone have a lead on where I need to go/what I need to do to acquire these secret barcodes?

    Thanks

The barcodes are on the pacb.com website

Click on the "Product Details+" below barcoded M13 primer plate. The barcode sequences are in the Primer Sequences (FASTA) link.

    Cheers,
    John

        Current Approaches to Protein Sequencing
        by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
