  • Does anybody have SOLiD v. 3.5 run protocol

    Hello, everyone.

I have already discribed our problem on this board. In short: we currently operate SOLiD 4, but still have a considerable amount of version 3.5 consumables, which we would really like to use.

The only difference between these two versions is the run protocol. It is a xml file (named something like SOLID3Opti.xml or similar) located by default in c:\protocols\ folder of Windows operated computer. It refers to fluidic subsystems script files (they have '.waa' extension) which are located by default in c:\flowcellscripts\ folder.

If anybody could provide me the content of these two folders we could use our v. 3.5. consumables. Thank you in advance.

you can run 3.5 chemistry on a solid4 without these changes. you just needed to add this : SOLiD™ Mixing Strip Tubes with Zinc.

      Thank you, snetmcom.

Have you personally tried this approach? For I was sure that SOLiD™ Mixing Strip Tubes with Zinc is only required for ver. 4 sequencing reagents, while ver 3.5 is self-sufficient.

I have managed to find the fluidics scipts, still in search for .xml run protocol ver 3.5 file.

        Current Approaches to Protein Sequencing
        by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

        Strategies for Sequencing Challenging Samples
        by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
