I have just started to work on solid 5500xl and planning to prepare a mate pair library 3kb.
I just need to know what would be the imp steps to be taken care during library preparation step.
The nick translation time given in the protocol is 10-11 mins. Since the nick translation time decides the length of the final library, what time of incubation should i keep (ie 10 or 11 mins.) Right now I am following the protocol as it is. I would also like to know the range of the final library, with the peak, as assessed on Bioanalyzer
Anybody tried increasing the nick translation time and seen the results.
Also what would be the maximum no. of amplifications that can be given.
