    SOLiD5500 (total) meltdown

    Hello,

    we are experiencing problems with our 2 SOLiD5500 machines since end of july. It is getting extremely frustrating because one run the reverse quality is crap (<40% good best), another one the forward is crap (<20% good best)and the next one the barcode is crap (<60% good best).

    Is there anyone else experiencing problems with their machines when comparing results to previous ones?

    Sidenote: we do see that lane 1 is routinely better than lane 6, although lifetech assured us this was impossible.

    Are you certain the library titration was correct? Best beads % can be low from too much DNA in bead prep.

      Yes I am sure the samples are good, we did run them previously without problems. The noise/signal and on axis are all ok.

      The problem does not appear to be the machine so now we are testing another EZbead station and another lot-nr chemistry.
