    Innovated clonal amplification

    Hi Colleagues,

I wonder what is the difference between Avalanche and Wildfire methods of generation clones from single molecules? They seem to be synonyms.

Sorry, I have forgotten that Wildfire is matched to Solid instruments and has been available for some time.

I realise that this is replying to a very old Thread, but my understanding is that Wildfire was the clonal amplification technology applied to SOLiD, whereas Avalanche was the (same) technology applied on the Ion Torrent platform.

From what I can see, it all went very quiet with these clonal amp technologies several years ago: have they died the death? I recall seeing some user* application of Wildfire on the SOLiD 5500W - with the W presumably being 'Wildfire'? - *Audrey Papp, The Ohio State University.

Hi - Any chance someone has raw images (i.e. tiffs) from a 5500W? Curious to learn more about cluster size, shape, intensities from template walking.

        Thanks!

Cluster shape and intensities were super.

Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.

I’m not a sequencing expert. I’m a purification scientist who uses NGS to evaluate workflows my group develops. With this perspective, we think about the sample first and the NGS workflow second. The sequencer is an exceptionally honest reporter, but it can only report on what you give it, so whether you get clean, interpretable data from an NGS workflow is largely determined before you begin.

Here are nine questions we think about, in roughly the order they matter, before...
