    ribosomal profiling on a SOLiD 5500?

    I'm starting a ribosomal profiling experiment where I want to do the sequencing on a SOLiD 5500xl. I've done some work with Illumina sequencing. Is there a protocol for making the libraries for the SOLiD? I think I understand that I need to use specific DNA/RNA adapters (Adapters A or B), but these do not seem to currently commercially available. Do i need to go ahead and synthesize them myself? Has anyone done this?

    Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.

    The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.
