Hello Everyone, we aim to study Circular RNA in a few days. Can you give advice for optimization and Library Prep phase? I will be using the Illumina Total RNA Ribo Zero Plus kit. My understanding is that to obtain circular RNA, I will need to add an extra cleanup and amplification step after the rRNA Depletion steps before creating First Strand DNA. Which enzyme should I use to obtain Circular RNA at that stage? Which one would you recommend? What exactly will the enzyme we use do? Does it have anything to do with the A-tailing stage?

Data variability is still an issue in sequencing technologies despite the advances in reproducibility and accuracy of these platforms. But the problem does not originate in the sequencing itself, but in the previous steps, before the sample reaches the sequencer.
The first step is collection, followed by preservation and sample preparation for analysis. Most scientists overlook those steps, but not being careful might just be skewing the experiment’s results.
