    Illumina adapter--what can I dispense with?

    Hi folks,


    For my project I want to alter the 5' illumina adapter sequence (aka "universal" adapter) to make the 3'-most 11 bases different from the 11 bases on the other side (the symmetry in the two adapters screws up my experiment). Assuming I add a custom primer for read #1, does anyone expect this to be a problem?

    Dear Uberfinch,
    This should not be a problem, unless you will be running paired-end reads. You are unable to use a custom seq.primer on read two since each lane is not addressed separately at that stage.

      Love this - good data definitely starts from good input, and poor input can only give relatively poor data. I particularly like the mention of Nanodrop/absorbance based methods for quantification. It's such a toss up if you'll get an accurate reading or what amounts to a randomly generated number, and a lot of library/sequencing related issues can be traced back to poor quant.
