Hi guys,
I have two queries about the homemade AMPure XP beads.
1) Almost in all the homemade recipe of AMPure XP beads, there mentioned the adding 0.05% Tween 20. However, as soon as you add the Tween 20 (or other detergents like Triton X-100, NP-40, SDS, etc.), the whole reagents get viscous. And when you try to transfer the reagents with micropipettor, it is hard to drain the entire liquid with one press, ie., it has residue at the top of the tips. But AMPure XP is not like that.
2) The pH value of homemade AMPure XP drops gradually, and gets to below 7.0 in about three months (the initial pH value is 8.0). While AMPure XP's pH is very stable at about 9.0.
Has that ever happed to you?
