  • Performing Hybrid Assemblies With Newbler

Has anyone performed assemblies with GSAssembler (ver. 2.0.01) using Sanger data (fasta) in combination with 454 data? A friend has done this for me with miraEST and I was going to try it with Newbler. Just wondering what opinions were as to the quality of similar assemblies.

Any particular reason for using Newbler gsAssembler v2.0.01 when v2.3 is out? As I recall, the main new feature is better support for EST assemblies.

Good point! I got this ver. from our core facility, so maybe they gave me the wrong install disk since this summer the Roche rep. said it was the latest "hot off the presses" version. I should be able to get the latest ver. So, have you done this type of assembly with the new version?

Could be - they can probably get the latest version from Roche for you if they haven't already got it themselves.

        No, not personally.

          Current Approaches to Protein Sequencing
          by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...

          Strategies for Sequencing Challenging Samples
          by seqadmin

Despite advancements in sequencing platforms and related sample preparation technologies, certain sample types continue to present significant challenges that can compromise sequencing results. Pedro Echave, Senior Manager of the Global Business Segment at Revvity, explained that the success of a sequencing experiment ultimately depends on the amount and integrity of the nucleic acid template (RNA or DNA) obtained from a sample. “The better the quality of the nucleic acid isolated...
