    Ribo-zero - FastQC plots - Per sequence GC content

    Dear all

    Running Ribo-zero kits on human data, seen consistent FastQC plots like this.
    Looks like a few very highly expressed ncRNAs, a result of the kit producing far more variable coverage and types of transcripts? Any comments appreciated.

    Hi Elsie, did you ever figure this out? We are seeing something very similar in some recent RNA-seq data generated using the Ribo-zero kit. We have never seen such things when we use poly A selection. We removed residual rRNA sequences (~2-6%) and that eliminated some of the weirdness, but the profile is still quite odd. David

      Even though the profile may be odd you should go ahead and analyze the data to see what it looks like. Things may be fine.
