  • RNA-seq libraries from bird blood

    Hi all,
    I am planning a RNASeq experiment and I would like to prepare some Illumina libraries. The starting material is avian blood. I had a look around but it is not clear which is the best method to isolate high quality RNA from this highly nucleated sample. Do you have experience or suggestions? What are the right procedures from the blood storage to the final isolation?
    Is depletion of hemoglobin mRNA necessary? Can be a problem in the evaluation of gene expression?

    I would use Trizol followed by RNAEasy. Use much less blood (like 5 to 10x less) than with mammalian blood.
