    Thermal cycler with variable temperature lid?

    Does this machine exist:, for example, a machine that can change the lid temperature during the course of the reactions? Something tells me that having the lid set to 75C when my reverse transcription reaction is occurring at 45C is not a good idea. In addition, I've been getting lots of adapter dimers and/or technical sequences/artifacts in library prep which I see on the bioanalyzer during my library constructions. Could not having a heat-changing lid be the reason? I am talking about cross all different sample types/library prep kits/depletions.

    I'm not aware of such a cycler. But having the lid temperature on 75°C seems to me very low. For all kind of reaction we use 105°C.

    But I'm unsure whether this can lead to your problem, as the higher lid temperature should just prevent vaporescence of your sample.
