  • a Great 454 Run

    Hi all,

I've made a great run, i will share my mods when then output is validated.

the throughput was more than 700MB and it's the regular FLX.

Yes, this is how far you can stretch GS FLX Titanium (we have seen the same on our instrument). Congrats!

These are amazing numbers! This is a shotgun library, presumably. What was your DNA:bead ratio and how did you determine that ratio?

          Thanks.

well I've targeted a 14% enrichment just to overcome the two oil cups and run the full ptp using one cup emulsion and everything was chilled. the library size or the fragmentation was from 900-2.5kb the majority was around 1.1-1.2kb.

yet still to validate, another run was performed with the same modifications and the throughput was 700+ too.

Essential Discoveries and Tools in Epitranscriptomics
                by seqadmin

The field of epigenetics has traditionally concentrated more on DNA and how changes like methylation and phosphorylation of histones impact gene expression and regulation. However, our increased understanding of RNA modifications and their importance in cellular processes has led to a rise in epitranscriptomics research. “Epitranscriptomics brings together the concepts of epigenetics and gene expression,” explained Adrien Leger, PhD, Principal Research Scientist on Modified Bases...

                Current Approaches to Protein Sequencing
                by seqadmin

Proteins are often described as the workhorses of the cell, and identifying their sequences is key to understanding their role in biological processes and disease. Currently, the most common technique used to determine protein sequences is mass spectrometry. While still a valuable tool, mass spectrometry faces several limitations and requires a highly experienced scientist familiar with the equipment to operate it. Additionally, other proteomic methods, like affinity assays, are constrained...
