  • Maximum # of copies in a cluster?

    Hi all,

    I am just wondering why the # of DNA copies in each cluster is capped between 1000-2000.

    Is this just due to the size of the well? I'm guessing that all fixed oligos in a well are extended during bridge amplification.

    Or does the sequencing process fail w/ too many copies?

    Only Illumina knows. I am guessing the isothermal clustering for the X10 and the HS4K is mostly limited by the real estate (and resulting steric problems for the bridged molecules?) and the number of flowcell oligos per nanowell.
