  • Opentrons Automated Pipetting System

Has anyone here used the Opentrons OT-2 for library preparation? https://opentrons.com/ot-2

"Accuracy Accurate down to 384-well plates"

This is a new measure of accuracy that I was previously unaware of.

Their only 'Sequencing Application' in their protocol library is for 16S. There are liquid handling platforms with a much wider array of NGS protocols ready to go than this..

A neighboring lab has the OT-One. This first version is unusable for just about anything - it can't hold the pipettes properly and the plates have a lot of play. Version two looks very different, though.

There have recently been a few discussions about them on Reddit; perhaps it might be a way to get in touch with people who use them.

We recently got an OT2. So far it works for single channel pipetting.

We have problems with both of our multi-channel pipettes.
The magnetic module seems to have a design flaw - the magnet is too close to the samples, even when disengaged and at maximum distance (achieved with an self-made adapter). Thus, the beads do not stay in suspension for long - ideally they should stay in suspension for 5 minutes before then engaging the magnet.

Understanding Genetic Influence on Infectious Disease
            by seqadmin

During the COVID-19 pandemic, scientists observed that while some individuals experienced severe illness when infected with SARS-CoV-2, others were barely affected. These disparities left researchers and clinicians wondering what causes the wide variations in response to viral infections and what role genetics plays.

Jean-Laurent Casanova, M.D., Ph.D., Professor at Rockefeller University, is a leading expert in this crossover between genetics and infectious...

Addressing Off-Target Effects in CRISPR Technologies
            by seqadmin

The first FDA-approved CRISPR-based therapy marked the transition of therapeutic gene editing from a dream to reality1. CRISPR technologies have streamlined gene editing, and CRISPR screens have become an important approach for identifying genes involved in disease processes2. This technique introduces targeted mutations across numerous genes, enabling large-scale identification of gene functions, interactions, and pathways3. Identifying the full range...
