Squeezing the most out of Nextera XT...?
Hi All,
I'm wondering if anyone has experimented with pushing the limits of Nextera XT in order to get more samples out of a kit? I want to sequence a bunch (~150) of mitogenomes generated via long-range PCR. I was hoping to find a way to stretch the Nextera XT kit (and indices) to get the most bang for my buck. Since the samples are amplicons I think I should have enough "room" on one MiSeq run to collect all 150 mitogenomes at high depth...but don't want to have to pay for 150 Nextera XT libraries. Any ideas? Can I just run smaller reactions of the Nextera XT sample prep kit?
