  • 454 Junior MIDs

    Hey folks.

    I'm attempting to do a 454 Junior run with 54 pooled libraries, and it seems that there are only 14 suggested MIDs for use with the GS Junior chemistry. Supposedly these are optimised for easy differentiation, but theoretically you can create your own additional tags as well. Does anybody have success with homebrewed MID tags and care to share some sequences? Or is there a way to combine multiple tags? Some of my amplicons could be slightly larger than the available read length, so putting an MID on each end might not work.

    Any feedback is appreciated!

    Extended MID List

    Here is the extended List of MIDs I reference when designing barcoded adaptors that are incorporated during PCR. I have only used the first 18 on this list and don't know how well any of the others work.

    These are not the MIDs for the RAPID ligation-library prep. Those are available in another Tech Bulletin and on IDTs website. I can dig up that list if required.

      Hi Newendophytologist,

      As sdl11 pointed out, there is an extended list of MIDs with an additional 120 MID adapters. These are in addition to the 12 MIDs provided in the GS Rapid Library MID Adapters Kit. More information on these Rapid Library MID Adapters is found in Technical Bulletin No. 2010-010 on our customer accessible website my454.com.
