    TruSeq small RNA for directional sequencing

    Has anybody tried the TruSeq small RNA sample prep kit to prepare libraries for directional mRNA sequencing? We would like to multiplex some bacterial samples, and this kit should work if we fragment the RNA first.

    Pair end adapter stripping

    I am using small RNA Truseq kit for pair end directional RNA seq. I am just wondering if somebody knows what adapters' sequence I can use to strip the 3' and 5' adapters. My fragments are shorter than the read length and it is important to strip off the two adapters. Thanks!

      Has anyone added the Parkhomchuck UNG step to TruSeq RNA?

        Yes. I've done about 20 samples using Parkhomchuck mods with TruSeq RNA reagents and it works pretty well. I get about 93% of the reads in the correct direction.
